Supporting data for manuscript "Characterizing vascular function in mouse models of Alzheimer’s disease, atherosclerosis and mixed Alzheimer’s and atherosclerosis"
The excel file 'animalIDsandGrps' contains the animal ID, sex, weight, and age of all animals for whom data was collected from for the experiments included in the manuscript "Characterizing vascular function in mouse models of Alzheimer’s disease, atherosclerosis, and mixed Alzheimer’s and atherosclerosis". Note that female mice were excluded from the final dataset due to 2/4 of the groups (ATH, MIX) not including females.
The CSV file 'EyreShawetal_MixedDisease_SourceData' contains the the averaged time series traces and extracted metrics from individual experiments used across Figures 2-6 in the present manuscript. The following acronyms included in the CSV file are defined: Hbt is total hemoglobin, Art is artery region, SEM is standard error mean, TS is time series, max peak is maximum peak, AUC is area under the curve, WT is wild-type, AD is Alzheimer's disease, ATH is atherosclerosis and MIX is mixed AD/atherosclerosis. The locomotion values (traces and metrics) are in arbitrary units with larger integers representing a greater displacement of the spherical treadmill, the hemodynamic (Hbt) values (traces and metrics) are a percentage change from the normalised baseline (prior to stimulus presentation), and the corresponding time series vector is presented in seconds.
The MATLAB files '2sStimData_indTrials' and '16sStimData_indTrials' contain the individual time series for HbT, HbO and HbR in the whisker, artery, vein and parenchyma ROIs for all collected trials (N=3479 for 2s stim paradigm, N=1155 for 16s stim paradigm). The accompanying locomotion traces and time vector are also included. These time series traces are 2-dimensional with dimension 1 (rows) representing the individual stimulation trials which the data has been cut around, and dimension 2 (length) representing the number of frames captured (200 frames, recording speed 7.9932 frames per second). The table T contains the labels for all individual trials: animal ID, group ID (i.e. 2=WT, 1=AD, 3=ATH or MIX), sex (1=male, 2=female), session ID (corresponding to the experimental imaging day), and trial ID (corresponding to the order of detected locomotion events within each experimental imaging session); as well as the extracted metrics (e.g. AUC, max peak, minima, time to peak).
The excel files '2sStim_data4LMM', '16sStim_data4LMM', and '2sStim_locoRank_4LMM' contain the time series metrics (including maximum peak, AUC etc) with corresponding labels (including animal ID, sex, session ID, etc) present in the MATLAB file extracted into spreadsheets for loading into RStudio to conduct statistical analyses using linear mixed models. The locoRank file is used for the alternative method for removing the confounds of locomotion used in Supplementary Figure 4.
